Product Information:

Alyftrek is indicated for the treatment of cystic fibrosis in patients aged 6 years and older with at least one F508del mutation in the CFTR gene, or another responsive mutation.

Alyftrek is a combination tablet containing vanzacaftor, tezacaftor, and deutivacaftor taken 1 time a day.

Your health care provider will have you undergo a blood test to determine if you carry the F508del mutation or another responsive mutation.
Alyftrek has a triple mechanism of action. First, vanzacaftor and tezacaftor work in tandem binding at different sites to the CFTR protein-on which cystic fibrosis results due to mutations-and thereby facilitate its trafficking to the surface of cells. Second, deutivacaftor enhances its function there. Finally, when all three drugs are used together, you get more working CFTR proteins at the cell surface. Consequently, chloride transport is improved, as measured in studies both in the laboratory and by assessments of chloride levels in the sweat of patients.

How do I take Alyftrek?

Alyftrek tablets are taken 1 time a day. You may have to take up to 3 tablets as a single dose.
Take Alyftrek tablets whole.
Take the doses at about the same times every day.
Take Alyftrek exactly as your healthcare provider tells you to take it. Take Alyftrek by mouth only.
Take Alyftrek tablets with food that contains fat. Examples of fat-containing foods include butter, oil, eggs, peanut butter, nuts, meat, and whole-milk dairy products such as whole milk, cheese, and yogurt.

Dosage of Alyftrek tablets for children aged 6 to <12 years, weighing less than 88 pounds [40 kg])
Take 3 tablets at the same time each day of the purple round-shaped tablet.
This is marked with ‘V4’, and contains the medicines vanzacaftor 4 mg, tezacaftor 20 mg, and deutivacaftor 50 mg.
Dosage of Alyftrek tablets for adults and children aged 6 to less than 12 years weighing 88 pounds [40 kg] or more, or 12 years and older):
Take 2 of the purple oblong-shaped tablet at the same time each day.

This is marked as ‘V10’, and it contains the medicines vanzacaftor 10 mg, tezacaftor 50 mg, and deutivacaftor 125 mg.

Side effects of Alyftrek:

Common Alyftrek side effects include:
cough
pain in the mouth or throat
pain or swelling of your nose or throat (nasopharyngitis)
flu (influenza)
tiredness
Including stuffy and runny nose, an upper respiratory tract infection or common cold an increase in liver enzymes rash headache congestion of the sinuses.
